DNA Encoded Libraries
To escape from flatland, natural product inspired novel building blocks for DNA encoded libraries are needed. For the inclusion in either linear, cyclic or branched libraries, the functional groups need to be separated by spirocycle or bicycle scaffolds which allows for incorporation of unique 3D structure and additional diversity points for attachment. These building blocks have MW from 150 to 250 Da. DEL synthesis is limited by the chemical reactions which include amide formation, reductive amination, Suzuki reaction, displacement of halides or copper catalyzed click reactions.
